Book excerpt: Describes the general approach of combining liquid chromatography with matrix-assisted laser desorption ionization mass spectrometry for on-line detection of peptides and proteins, based on the use of a continuous flow probe. Recent advances in time-of-flight mass spectrometric instrumentation for this approach are reported. Finally, the results of time-lag focusing matrix-assisted laser desorption ionization analysis of 15 peptides and proteins using solid matrices are presented. Mass measurement error is discussed along with the limitations of, and potential improvements to, the analytical system proposed.
